In this study, biguanide-containing tetrazole compounds was synthesised from the reaction of guanidine and NaN3. The structure of the compound was characterised by spectroscopic and analytical methods. Single crystals of the compounds were obtained and the solid-state structure was characterised by X-ray diffraction study. DNA binding affinities (binding coefficients) and binding modes of the synthesised compounds were determined by spectroscopic and analytical methods. The effects of the compounds on DNA binding affinities and their comparison with standard DNA binding agents. The compounds were found to have a binding constant close to ethidium bromide (4 × 105 M−1). In addition, the effects of A and B molecules on lung cancer cell lines (A549, Calu1, and H1650) and bone cancer cell line (Saos2) were investigated and the TGI values between 321.61 – 219.99 µM and LC50 values between 408.03 - 1112.14 µM against only H1650 cell line showed much higher anticancer performance than the control anticancer drug 5FU. The fact that the TGI values of molecules A and B (458.30 - >1600 µM) are higher in normal cells than in cancer cells shows that both can be used safely.
