In this study, determination of total protein and pigment amounts of Scenedesmus bernardii G.M. Smith which was
isolated from Yeşilırmak (Kat Town-Tokat) and Cladophora fracta (Dillw.) Kuetzing which was isolated
from GOÜ Campus Pool was aimed. After Scenedesmus bernardii was cultivated in Bristol growth
medium, total protein contents were observed with Bradford method. Cladophora fracta was exposed to
protein analysis after being isolated without being reproduced in growth medium. Both algae samples
were homogenized with phosphate buffer after being fragmentized in liquid nitrogen. Furthermore,
Coomaasie brillant-blue G-250 added to supernatant and their absorbance values in 595 nm was
determined after an incubation of 10 minutes. As for pigment analysis, algae were again fragmentized
in liquid nitrogen and absorbance values in 450, 645, 663 nm in spectrophotometer were observed for
chlorophyll and carotenoid amounts of the algae homogenized in acetone of 80 %. Total protein
contents at dry weights of algae were calculated by drawing calibration curves above absorbance value
of the standard (bovine serum albumin). According to this, total protein amounts were found as 0,209
mg/g (20.9 %) in Cladophora fracta; while it is 0,241 mg/g (24.1 %) in Scenedesmus bernardii.
Chlorophyll and carotenoid amounts were calculated with respect to Arnon and Jaspars equations.
Chlorophyll and carotenoid amounts were determined as follows in Cladophora fracta; chlorophyll-a as
0,364 mg/g (36.4 %), chlorophyll-b as 0,208 mg/g (20.8 %), total chlorophyll amount as 0,572 mg/g; as
carotenoid amount 0,129 mg/g (12.9 %). While they were determined as follows in Scenedesmus
bernardii; chlorophyll-a as 0,677 mg/g (67.7 %), chlorophyll-b as 0,328 mg/g (32.8 %) total chlorophyll
amount as 1,00 mg/g and carotenoid amount as 0,186 mg/g (18.6 %).
