Evaluation of pannexin localisation in mouse ovarian and testis tissue

Özet

Objective: Cell-to-cell and cell-to-matrix communication is crucial for the development of ovarian follicles, the maturation of oocytes, and the development of spermatogenic cell lines. Our study aimed to investigate the zof the Pannexin (Panx) family members; Pannexin 1 (Panx1), Pannexin 2 (Panx2), and Pannexin 3 (Panx3) in mouse ovarian follicles and in each component of the testis tissue spermatogenic cells.

Methods: Pannexins were localised in primordial, primary, secondary and tertiary follicles in ovarian tissue and in the spermatogenic cells in the seminiferous tubule in testis tissue and their severity was analysed by the immunohistochemical method.

Results: Immunohistochemical analysis revealed no significant difference in the intensity of Panx1 expression in follicle cells and follicular oocytes. However, the signal intensity of Panx2 and Panx3 was higher in follicular oocytes and cells (p<0.05). In spermatogenic cells, there was no difference in Panx1 intensity, whereas Panx2 and Panx3 proteins were intensely expressed especially in spermatogonium and primary spermatocytes (p<0.05). Furthermore, Panx2 was found to be concentrated in the membrane area of Sertoli cells in the seminiferous tubule. In the germinal epithelium and adipose tissue, all Panx members were found to be densely localized in both ovarian and testis tissues.

Conclusion: This study provides in vitro evidence suggesting that Panxs play an important role in the communication of mice's ovarian follicles and their oocytes, as well as male germ cells.
