5S and 18-5.8-26S nuclear ribosomal DNA (rDNA) units are located in most eukaryotes. 5S and 18-
5.8-26S rDNA occur at one or several specific loci and rDNA are highly conserved repetitive
sequences. Polymorphism or conservatism of rDNA copy number and chromosomal localization are
comparable and viewable. Accordingly ribosomal DNA units are used on interspecific phylogenetic
relotionships. Additionaly rDNAs are preferred and useful cytogenetics markers for chromosome
analysis. Several agricultarly important cultivated plants which are wheat, barley, rice, cotton etc. are
worked for understanding this concept. Sainfoin is a member of fabacaea family which is
agriculturally important family. Sainfoin is a fodder crop that is preferred because of not bloating to
animals. Also nectar-rich sainfoin has nitrogen fixing capacity like other fabacaea members. Sainfoin
is dry and cold resistant and can grow in arid and chalky areas. These characteristics makes it
important option for breeding studies. Making sense of genome organization and phylogenetic
analysis a cultivar illuminates breeding process. Considering this information, visualization of 45S
rDNA and 5S rDNA genes by FISH on sainfoin is studied. In research, Onobrychis viciifolia (sainfoin),
Özerbey variety are used for plant material. Research results shows, number of 45S rDNA locus is 6
and the number of 5S rDNA locus is 8 in interphase.
