Abstract

The Onobrychis genus belongs to the Hedysareae tribe within the Fabaceae family. The genus includes species with basic chromosome numbers of seven or eight, existing as diploid (2n = 2x = 14, 2n = 2x = 16), tetraploid (2n = 4x = 28, 2n = 4x = 32) or octoploid (2n = 8x = 56) species ¹. The sainfoin needs to be improved in terms of genetic studies and has great potential for agriculture. Therefore, comparative genetic information about the Onobrychis genus is valuable for germplasm use in breeding. CenH3 is a variant of histone 3 protein and is involved in the assembly of the kinetochore. It is essential for phylogenetic analysis and elucidating the evolutionary process of the centromere ³. An earlier study recognized five different CenH3 genomic loci in O. viciifolia ² .Parallel to this, the genomic sequences of the CenH3 genes in a set of wild species, O. altissima, O. biebersteinii, O. hajastanaand O.oxydanta are determined by molecular cloning in our study. Using gene-specific primers, PCR fragmentation was detected between 4 and 7 in wild species. A detailed analysis of the results will be included in our poster. Our research, presented here, will contribute to the comparative analysis of both cultivated and wild-related species with unique outputs. Additionally, we will discuss the phylogeny and evolutionary conservation of the CenH3 gene in the Onobrychis genus.
